Tune all six strings cleanly
Mute the other five strings while the tuner listens
Let the low B0 settle after the initial pluck transient
Check high C3 against G2 at the fifth fret
Tune upward into every target and make a second pass
Check the octave, not only the letter
Standard six-string bass uses B0 at about 30.87 Hz and C3 at about 130.81 Hz. B1 or C2 is the wrong octave even though the note letter matches.

History & context
B E A D G C extends standard four-string bass equally in both directions: one lower string for B0 and one higher string for C3. The uniform fourths preserve familiar interval shapes across the neck.
